Enhancement of Endothelialization by Topographical Features Is Mediated by PTP1B-Dependent Endothelial Adherens Junctions Remodeling

Endothelial Cells (ECs) form cohesive cellular lining of the vasculature and play essential roles in both developmental processes pathological conditions. Collective migration proliferation endothelial cells are key underlying endothelialization vessels as well vascular graft, but complex interplay mechanical biochemical signals regulating these still not fully elucidated. While surface topography modifications have been used to enhance vitro, thus far such single-modality met with limited success. As combination therapy that utilizes multiple modalities has shown improvement addressing various intractable biomedical conditions, here, we explore a combined strategy topographical features conjunction pharmacological perturbations. We characterized EC behaviors response micrometer-scale grating concert perturbations adherens junctions (EAJ) regulators. found protein tyrosine phosphatase, PTP1B, serves potent regulator EAJ stability, PTP1B inhibition synergizing topographies modulate rearrangement, thereby augmenting global monolayer sheet orientation, proliferation, connectivity, collective cell migration. Our data delineates crosstalk between cell–ECM sensing cell–cell junction integrity maintenance suggests use inhibitor could be promising for promoting proliferation.
